Effect of Fuxian Qiangxin Recipe on sST2, CaN in chronic heart failure rats with yang deficiency and water flooding syndrome
Objective,2,To observe the effect of Fuxian Qiangxin Recipe on related indexes of myocardial remodeling in rats with chronic heart failure (CHF) induced by adriamycin, and to explore the intervention effect of Fuxian Qiangxin Recipe on heart failure.,Methods,2,Sixty SPF male Wistar rats were randomly divided into 6 groups: normal group, model group, captopril group, and Fuxian Qiangxin Recipe low, medium and high dose group, with 10 rats in each group. Except for the normal group, other groups were treated with intraperitoneal injection of adriamycin to replicate chronic heart failure models, and then given corresponding intervention measures. After 6 weeks, the general conditions of the rats in each group were observed. The morphological changes of myocardium were compared by HE staining. The cardiac function of the rats in each group was detected by cardiac color Doppler ultrasound. Enzyme-linked immunosorbent assay (ELISA) was used to detect the content of soluble matrix lysin-2 (sST2) in rat serums, and Western blot was used to detect the protein expression of calcineurin (CaN) in rat myocardial tissues.,Results,2,① General condition: Compared with the normal group, the rats in the model group showed symptoms of yang deficiency, such as obvious weight loss, less activity, dull hair, dim nose and mouth, excessive urination and thin stool. The symptoms of each treatment groups were improved compared with those of the model group. ② Pathological observation: The myocardial nucleus of the model group was more stained than that of the normal group, showing local vacuolation, indicating cardiomyocyte injury; compared with the model group, the degree of cardiomyocyte injury in each dose group of Fuxian Qiangxin Recipe was alleviated and improved in varying degrees. ③ Cardiac function: compared with the normal group, the left ventricular ejection fraction (LVEF) decreased, the end-systolic diameter (LVIDs) and end-diastolic diameter (LVIDd) increased significantly in the model group (,P,<,0.05), and the LVEF increased significantly and LVIDs decreased significantly in each group of Fuxian Qiangxin Recipe compared with the model group (,P,<,0.05). ④ Detection of sST2 and CaN: Compared with the normal group, the levels of serum sST2 and myocardial tissue CaN in the model group were significantly higher than those in the normal group, while the levels of sST2 and CaN in each dose group of Fuxian Qiangxin Recipe were lower than those in the model group (,P,<,0.05).,Conclusion,2,Fuxian Qiangxin Recipe can protect myocardium, improve cardiac function and delay myocardial remodeling in rats with heart failure. The mechanism may be related to the inhibition of serum sST2 and myocardial tissue CaN expression.
