Experimental study on expression of inflammatory factor protein and mRNA in plaque tissues of diabetic atherosclerotic rats treating with Zhige Tongmai Granules
Objective,2,To observe the effects of different doses of Zhige Tongmai Granules (ZGTMG) on the expression of protein and mRNA of tumor necrosis factor-α (TNF-α), matrix metalloproteinase-9 (MMP-9), vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule-1 (ICAM-1) and tissue inhibitor of matrix metalloproteinase (TIMP) in plaque tissues of diabetic atherosclerosis (AS) rats.,Methods,2,Ten of 70 SPF-grade Wistar male rats at 4 weeks of age were randomly selected as the normal group and administered with basic feeds only, and the remaining 60 were the diabetic modeling group. The diabetic atherosclerotic rat model was designed by streptozotocin (STZ) injection (30 mg/kg), intra-aortic balloon pump injuries and the high-fat diet. The modeled rats were randomly divided into a model group, ZGTMG low-dose group (low-dose group, gavage administration of ZGTMG at 1.05 g·kg,-1,·d,-1,), ZGTMG medium-dose group (medium-dose group, gavage administration of ZGTMG at 2.1 g·kg,-1,·d,-1,), ZGTMG high-dose group (high-dose group, gavage administration of ZGTMG at 4.2 g·kg,-1,·d,-1,) and rosuvastatin sodium group (modern medicine group, gavage administration of rosuvastatin sodium at 1.05 mg·kg,-1,·d,-1,) according to the blood glucose level, with 10 animals in each group. After 6 weeks of administration, atheromatous plaques were obtained from the abdominal aorta as the specimen and the protein and mRNA expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP were detected.,Results,2,The protein and mRNA expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP were lowered significantly in the high-dose group and modern medicine group compared with those in the model group, and the differences were statistically significant (,P,<,0.05). The ,TNF,-,α, mRNA expression, and the protein and mRNA expression levels of MMP-9, VCAM-1, ICAM-1 and TIMP were lowered in the low-dose and medium-dose groups compared with those in the model group, and the differences were statistically significant (,P,<,0.05). Significantly lowered protein expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP as well as the lowered ,TIMP, mRNA expression were detected in the high-dose group, and compared with the protein and mRNA expression levels in the low dose group the differences were statistically significant (,P,<,0.05). The protein expression levels of VCAM-1 and TIMP were significantly lowered in the medium-dose group compared with those in the low-dose group, and the differences were statistically significant (,P,<,0.05). The expression levels of MMP-9, ICAM-1 and TIMP were significantly lowered in the high-dose group compared with those in the medium-dose group, and the differences were statistically significant (,P,<,0.05).,Conclusion,2,ZGTMG can significantly lower the protein and mRNA expression levels of TNF-α, MMP-9, VCAM-1, ICAM-1 and TIMP in the plaque tissues of diabetic AS rats, decrease the expression of inflammatory factors and improve the inflammatory response of vascular endothelium, and the clinical effect of the high-dose group is better than that of the medium-dose and low-dose groups.
