Simultaneous determination of five constituents in Yinban Prescription by HPLC wavelength switching method
Objective,2,To develop an HPLC wavelength switching method for determining the contents of (R,S) -goitrin, chlorogenic acid, albiflorin, forsythoside A, irisflorentin in Yinban Prescription.,Methods,2,The separation was performed on an Agilent InfinityLab Poroshell 120 SB-C18 (3.0 mm×150 mm, 2.7 μm) at 35 ℃ using a gradient elution with 0.1% phosphoric acid acetonitrile and 0.1% phosphoric acid water. The flow rate was 0.8 mL/min and the detection wavelength was set at 240 nm within 0-4 min, 325 nm within 4-6.5 min, 230 nm within 6.5-9 min, 328 nm within 9-21 min, 265 nm within 21-24 min.,Results,2,(R,S)-goitrin, chlorogenic acid, albiflorin, forsythoside A, risflorentin had a good linear relationship with the peak area in the range of 0.145-18.560 (,r,=1) , 3.566-228.200 (,r,=1) , 2.281-292.000 (,r,=1) , 6.312-808.000 (,r,=1) , 0.238-30.480 (,r,=1) g, respectively. The average recoveries were 97.86%, 102.09%, 99.06%, 98.63%, 101.51%, with ,RSD, 1.90%, 1.43%, 1.62%, 1.40%, 1.94% (,n,=9), respectively.,Conclusion,2,The HPLC wavelength switching method is accurate, reliable and reproducible for quality evaluation of Yinban Prescription.
