Effect of Gangyusan on wound healing, serum inflammatory factors and collagen expression after hemorrhoid operation in rats
Objective,2,To explore the influence of Gangyusan on serum inflammatory factors TNF- α, IL-6, expression of wound regeneration collagen and wound histomorphology in rats after hemorrhoid operation.,Methods,2,Sixty SPF SD rats were randomly divided into blank control group, model control group, hemorrhoid lotion group, and low, medium and high dose groups of Gangyusan (,n,=10). Except for the blank control group, the rats in other groups were injected with 75% glacial acetic acid around the anus to establish hemorrhoid operation model. After successful modeling, the blank control group and the model control group were given fumigation with high pressure sterilization water . The hemorrhoid lotion group and the low, medium and high dose groups of Gangyusan were given corresponding drug fumigation and washing treatment. After treatment, the wound area was measured, the healing rate calculated, and the color and exudation of the wound observed with naked eyes at specific time points (1st, 3rd, 7th and 11th day). Inflammatory reactions such as inflammatory cell infiltration, fibroblasts, granulation tissue proliferation and tissue healing were observed under light microscope. After 10 days of continuous treatment, the histopathology of granulation tissue in each group was compared; the contents of IL-6 and TNF-α in serum and the expression of ,IL,-6 and ,TNF,-α mRNA in wound tissue were detected by enzyme linked immunosorbent assay (ELISA) and fluorescence quantitative assay. The expression levels of COL1 and COL3 in wound tissue were detected by Western blot method and immunohistochemical method.,Results,2,Compared with the model control group, Gangyusan significantly reduced the expression of ,IL,-6 and ,TNF,- α mRNA in wound tissue and the content of inflammatory factors IL-6 and TNF-α in serum, while the protein expression of COL1 and COL3 in wound tissue increased significantly (,P,<,0.05). Under light microscope, the formation of fresh granulation tissue, neat arrangement of fibroblasts and abundant new capillaries were seen in each treatment group, which were better than those in the model control group.,Conclusion,2,Gangyusan can reduce the expression of inflammatory factors such as IL-6 and TNF-α in serum and wound tissue of rats, promote the formation of granulation tissue, capillaries and fibroblasts, and promote wound healing after hemorrhoid operation in rats.
