Effect of acupuncture regulating Klotho expression on proteinuria and renal pathology in SHR rats
Objective,2,To observe the effect of acupuncture on proteinuria and renal pathology in spontaneous hypertension rats (SHR).,Methods,2,SHR rats were randomly divided into model group, losartan group, Shenshu acupuncture group, Geshu acupuncture group, and Shenshu + Geshu group, with 10 rats in each group, and 10 WKY rats were set as normal group. The losartan group was intragastrically administered with losartan tablets which were dissolved in normal saline, with 30 mg/kg once a day. The acupuncture groups were given with electro-acupuncture intervention according to the corresponding acupoints every other day. The model group was fixed like the acupuncture groups done and without acupuncture. All groups were intervened continuously for 12 weeks. 24-hour urinary protein was detected by biochemical method before intervention and at the end of 4th, 8th and 12th week of intervention. At the end of 12th week, kidney samples were taken to observe the pathological changes of renal ultrastructure by light microscope and electron microscope, and the positive expression area of Klotho protein in renal tissue was detected by immunohistochemistry.,Results,2,After intervention, the 24 h urinary protein in the losartan group and the acupuncture groups were lower than that in the model group(,P,<, 0.05). The renal pathological injury in the losartan group and the acupuncture groups were alleviated in varying degrees, and the expression of Klotho positive area in the losartan group and the acupuncture groups were higher than that in the model group (,P,<,0.05).,Conclusion,2,Acupuncture can regulate the expression of Klotho in kidney, reduce proteinuria and renal pathological damage in SHR rats, and delay the progression of hypertensive renal damage.
