摘要

急性肝损伤的治疗药物筛选与病理机制研究,很大程度上依赖于实验动物模型的建立和应用。选择目前研究常用的四氯化碳(CCl,4,)、α-萘异硫氰酸酯(ANIT)、3,5-二乙氧基羰基-1,4-二氢-2,4,6-三甲基吡啶(DDC)、D-氨基半乳糖/脂多糖(D-GalN/LPS)、对乙酰氨基酚(APAP)诱导的5种急性肝损伤模型,从模型制备、生化特点、病理机制、模型优势与不足、模型在中药药理研究中的应用策略等方面详细论述,希望为急性肝损伤防治的基础研究提供一定的借鉴。

Abstract

Drug screening and pathological mechanism of acute liver injury depend largely on the establishment and application of experimental animal models. In this paper,five common animal models of acute liver injury,including CCl,4,ANIT,DDC,D-GalN/LPS and APAP,were selected and discussed in detail from the aspects of model preparation,biochemical characteristics,pathological mechanism,advantages/disadvantages,application of traditional Chinese medicine pharmacological research,etc.,which is of great significance for the prevention and treatment of acute liver injury.
