摘要

目的：探讨大黄-丹参通过miRNA-155对脓毒症大鼠血清中炎症因子水平的影响。方法：Wistar大鼠分为正常对照组、脓毒症模型组、大黄-丹参中药组、miRNA-155抑制剂组,每组15只。除正常组大鼠外,其余大鼠建立脓毒症模型,在此基础上给予相应的药物干预。在造模3 h后检测大鼠血清中 IL-6、IL-10水平；应用RT-PCR检测大鼠血清中miRNA-155的表达。结果：脓毒症模型组大鼠血清 miRNA-155表达、IL-6和IL-10水平较正常对照组高(P<0.05),大黄-丹参中药组和miRNA-155抑制剂组的miRNA-155表达、IL-6和IL-10水平均较脓毒症模型组低(P<0.05),大黄-丹参中药组和miRNA-155抑制剂组的miRNA-155表达、IL-6和IL-10水平差异无统计学意义(P>0.05)。结论：大黄-丹参或可通过miRNA-155降低脓毒症大鼠血清IL-6、IL-10水平,从而减轻脓毒症炎症反应。

Abstract

Objective:To explore the effects of Radix et Rhizoma Rhei and Radix Salviae Miltiorrhizae on the levels of inflammatory factors in serum of septic rats by miRNA-155. MethodsWistar rats were divided into normal control group, sepsis model group, Chinese medicine (Radix et Rhizoma Rhei and Radix Salviae Miltiorrhizae) group and miRNA-155 inhibitor group, 15 rats in each group. Except the normal group, sepsis model was established with rats in the other groups and drugs were given accordingly. At 3 hours after modelling, the levels of IL-6 and IL-10 in serum were detected, and the expression of miRNA-155 in serum was detected by RT-PCR. Results:The expression of miRNA-155 and the levels of IL-6 and IL-10 were higher in the model group than in the normal control group (P<0.05). The expression of miRNA-155 and the levels of IL-6 and IL-10 were lower in the Chinese medicine group and the miRNA-155 inhibitor group than in the model group (P<0.05). There were no significant differences in the expression of miRNA-155 or the levels of IL-6 and IL-10 between the Chinese medicine group and the miRNA-155 inhibitor group (P>0.05). Conclusion:Radix et Rhizoma Rhei and Radix Salviae Miltiorrhizae may reduce the levels of IL-6 and IL-10 in serum of septic rats through miRNA-155, thereby alleviating the inflammatory reactions of sepsis.
