Effects of Wangshi Ganshu patch on rats with liver-depression and blood-stasis syndrome and those with acute hepatic injury
Objective:To evaluate the effects of Wangshi Ganshu patch on hemorheological indexes and platelet aggregation rate in the rat model of liver-depression and blood-stasis syndrome, and the protective effect for acute liver injury induced by CCl,4, in rats. Methods①=1\*GB3Wistar rats were randomly assigned into 5 groups, namely blank control group, liver-depression model group and the Wangshi Ganshu patch low-dose group (crude drug 0.96 g/kg), medium-dose group (1.92 g/kg) and high-dose group (2.88 g/kg), 8 rats in each group. Excluding the blank control group, rat model of liver-depression and blood-stasis was established using the "clip tail" method. Each group of rats was given the corresponding intervention. 14 days later, blood was collected to test the blood viscosity, blood reductive viscosity, erythrocyte rigidity index, erythrocyte deformation index and rate of platelet aggregation. ②=2\*GB3SD rats were randomly assigned into 5 groups, namely blank control group, hepatic injury model group, Wangshi Ganshu patch low-dose group (0.96 g/kg), medium-dose group (1.92 g/kg) and high-dose group (2.88 g/kg), 5 males and 5 females in each group. Excluding the blank control group, CCl,4, was used to induce acute hepatic injury model of rats. Each group of rats received corresponding intervention and 14 days later, related biochemical indicators of liver disease were determined. Results:①=1\*GB3Compared with the liver-depression model group, in the high-dose group, increase in the low-cut blood viscosity and blood reductive viscosity, middle-cut blood viscosity and blood reductive viscosity as well as high-cut blood viscosity and blood reductive viscosity was significantly lowered in rat model of liver-depression and blood-stasis (P,<,0.05); and the erythrocyte rigidity index, erythrocyte deformation index and erythrocyte aggregation rate were reduced significantly (P,<,0.05). ②=2\*GB3Compared with the liver-depression model group, in the middle-dose and high-dose groups, multiple biochemical indicators of acute liver injury induced by CCl,4, were lowered significantly (P,<,0.05). Conclusion:Wangshi Ganshu patch has certain therapeutic effect in treating diseases with liver-depression and blood-stasis syndrome and shows protective effect on liver damage caused by chemical substances.
