Effect of Fu-Fang-Li-Shao Pill on pro-inflammatory factors in blood of rats with chronic migraine induced by nitroglycerin
Objective:To explore the pharmacological mechanism of Fu-Fang-Li-Shao Pill (FFLSP) improving chronic migraine of the liver-yang hyperactivity type by observing changes in the levels of pro-inflammatory factors in plasma of rats with nitroglycerin-induced chronic migraine. Methods36 male Sprague-Dawley rats were randomly assigned into 6 groups:normal control (NC) group, migraine group, low-dose FFLSP (FFLSP-L) group, middle-dose FFLSP (FFLSP-M) group, high-dose FFLSP (FFLSP-H) group and flunarizine hydrochloride (FH) group. FFLSP at corresponding doses (420,0 and 1,0 mg/kg) was given to the FFLSP-L, FFLSP-M and FFLSP-H groups; FH (1 mg/kg) was given by gavage to the FH group, and an equal volume of 0.9% NaCl solution was given by gavage to the NC group and the migraine group. Each group was dosed once a day for 30 days. Except the NC group, migraine model was established by subcutaneous injection of nitroglycerin (10 mg/kg) to rats in the other 5 groups on days 18,1, 24,7 and 30 during treatment. The gene and protein levels of pro-inflammatory factors and 5-HT in plasma of rats in each group were detected with qRT-PCR, ELISA and Griess reagent. Results:①Compared with the NC group, the expression of iNOS, IL-1β, IL-6 and TNF-α mRNA in plasma increased significantly in the migraine group (P<0.001); the content of iNOS, IL-1β, IL-6 and TNF-α mRNA in plasma decreased significantly after FFLSP treatment (P<0.001). ②Compared with the NC group, the levels of IL-1β, IL-6, TNF-α and NO products in serum increased significantly in the migraine group (P<0.05); and they were reduced significantly after FFLSP treatment (P<0.05). ③Compared with the NC group, the level of 5-HT in serum decreased in the migraine group (P<0.05); and it increased significantly after FFLSP treatment (P<0.05). Conclusion:FFLSP may reduce inflammatory reactions and relieve migraine of the liver-yang hyperactivity type by reducing the gene and protein levels of pro-inflammatory factors in the blood.
