Anti-inflammatory and analgesic effects of Daphnis Cortex enteric-coated preparations on adjuvant arthritis rats
Objective:To investigate the anti-inflammatory and analgesic effects of Daphnis Cortex enteric-coated preparation on adjuvant arthritis rats. MethodsThe rat model of adjuvant arthritis was established by intradermal injection of Freund’s complete adjuvant. The rats were divided into the control group,model group,prednisone group,and low,middle and high dose groups of Chinese medicine, 12 rats in each group. The prednisone group was given prednisone 3.2 mg/kg by gavage,and the low,middle and high dose groups of Chinese medicine were given the medicine 0.396 g/kg,0.792g/kg,1.584 g / kg,respectively by gavage. The control group and the model group were given the same volume of 7.5% CMC-Na solution. After continuous intervention for 12 days,the swelling rate of voix pedis,the 50% mechanical withdrawal threshold (MWT) and thermal withdrawal threshold (TWT) were calculated on the 7th and 12th day after modeling. The serum levels of interleukin-6 (IL-6),prostaglandin E,2, (PGE,2,) and tumor necrosis factor- α (TNF- α) were measured by ELISA 24 hours after the last administration. The pathological changes were observed by HE staining in the knee and ankle joints of the rats. Results:①Compared with the control group,the swelling rate of voix pedis in the model group was higher than that in the model group (P,<,0.01) and the value of 50%MWT,TWT was decreased (P,<,0.01) but the levels of IL-6,PGE,2,TNF- α in serum were decreased (P,<,0.01). ② Compared with the model group,the swelling rate of voix pedis was decreased (P,<,0.01),the 50% MWT and TWT were increased (P,<,0.01),and the levels of the serum IL-6,PGE,2,TNF- α were decreased in the prednisone group,and in the low,middle and high dose groups of Chinese medicine (P,<,0.01). ③ There was no significant difference among prednisone group and Chinese medicine groups in the above indicators (P,>,0.05). Conclusion:Daphne Cortex enteric-coated preparation has obvious anti-inflammatory and analgesic effects. It can significantly improve the pathological damage of knee joint and ankle joint in rats with adjuvant arthritis.
