Therapeutic effects of echinacoside on lead acetate induced dyszoospermia in mice
Objective:To discuss the therapeutic effects of echinacoside on lead acetate induced dyszoospermia in mice. Methods60 male BALB/c mice were randomly divided into the control group,model group,positive drug group and echinacoside groups with low-,middle- and high-dose,10 mice in each group. Except the control group,the mouse model of dyszoospermia was established by intraperitoneal injection of lead acetate at daily dose of 20 mg/kg for continuous 7 days in the other groups. After successful modeling,the positive drug group was treated with human chorionic gonadotropin(HCG)by intraperitoneal injection at daily dose of 500 U/kg. The echinacoside groups with low-,middle- and high-dose were treated with echinacoside by intragastric administration at doses of 25 mg/kg,50 mg/kg and 100 mg /kg,respectively. The control group and the model group were treated with 0.9% NaCl solution by intragastric administration at equivalent volume,with a course of 30 days. 24 hours after the last intragastric administration,the epididymis was separated,the sperm density,sperm motility rate and teratospermia rate were observed. The testicular tissue was fixed to observe the histopathological changes. The serum was separated and the testicular tissue was homogenized,the level of testosterone(T)in testicular tissue and the levels of luteinizing hormone(LH)and follicle stimulating hormone(FSH)in serum were detected by enzyme linked immunosorbent assay. Results:Compared with the control group,the sperm density and the sperm motility rate were decreased and the teratospermia rate was increased in the model group,and the T level in the testicular tissue homogenate was decreased and the levels of FSH and LH in serum were increased,with statistically significant differences(P<0.01). Compared with the model group,the sperm density and the sperm motility rate were increased and the teratospermia rate was decreased in the echinacoside groups with different doses,and the levels of FSH and LH in serum were decreased and the T level in the testicular tissue homogenate was increased,with statistically significant differences(P<0.01). The therapeutic effects of echinacoside on the reproductive impairment were in a dose-dependent manner. The histological observation showed that the pathological injuries in the testicular tissue were improved in echinacoside groups with different doses. Conclusion:Echinacoside shows obvious therapeutic effects on lead acetate induced dyszoospermia in mice,and its mechanisms may be associated with improving the reproductive hormone levels and recovering the regulation function of hypothalamus-pituitary-gonad axis.
