目的:观察荆防败毒散对阿霉素诱导肾病大鼠肾组织足细胞Nephrin、CD2相关蛋白(CD2AP)、整合素连接激酶(ILK)蛋白及基因表达的影响。 方法:SD大鼠随机分为正常组、模型组、苯那普利组、荆防败毒散组。除正常组外,其余各组采用大鼠尾静脉一次性注射阿霉素制备肾病模型,并进行相应干预。检测各组大鼠24 h尿蛋白水平及肾功能情况,检测肾组织中Nephrin、CD2AP及ILK蛋白及基因表达情况。 结果:与正常组比较,模型组大鼠24 h尿蛋白水平显著上升,肾功能指标增高,肾组织中Nephrin、CD2AP蛋白及基因表达明显降低,ILK蛋白及基因表达显著上调。与模型组比较,荆防败毒散组大鼠24 h尿蛋白水平显著降低,肾功能指标下调,肾组织中Nephrin、CD2AP蛋白及基因表达明显上调,ILK蛋白及基因表达显著下调。 结论:荆防败毒散可减少阿霉素肾病大鼠蛋白尿水平,其可能机制是下调ILK 表达,从而上调足细胞裂孔隔膜蛋白 Nephrin、CD2AP表达。
Objective:To observe the effects of Jingfang Baidu Powder on the protein and gene expressions of Nephrin, CD2-associated protein (CD2AP) and integrin linked kinase (ILK) in podocytes of adriamycin-induced nephropathy rat renal tissue. MethodsSD rats were randomly divided into the normal group, model group, benazepril group and Jingfang Baidu Powder group. Except the normal group, the rat model of nephropathy was established in other groups by single intravenous injection of adriamycin in tail. Each group was treated with the corresponding drug. The levels of 24 h urinary protein and renal function indexes in rats of each group were detected. The protein and gene expressions of Nephrin, CD2AP and ILK in the renal tissue were detected. Results:Compared with the normal group, the levels of 24 h urinary protein and renal function indexes were obviously increased in the model group, the protein and gene expression levels of Nephrin and CD2AP were obviously down-regulated, whereas the protein and gene expression levels of ILK were significantly up-regulated in the model group. Compared with the model group, the levels of 24 h urinary protein and renal function indexes were obviously decreased in the Jingfang Baidu Powder group, the protein and gene expression levels of Nephrin and CD2AP were obviously up-regulated, whereas the protein and gene expression levels of ILK were significantly down-regulated in the Jingfang Baidu Powder group. Conclusion:Jingfang Baidu Powder can decrease the proteinuria level in adriamycin-induced nephropathy rats. Its mechanism may be associated with the down-regulation of ILK expression and the up-regulation of Nephrin and CD2AP expressions in podocytes.
