Effects of curcumin combined with cisplatin on expressions of Bcl-2 and Caspase-3 in lung cancer A549 cells
Objective:To discuss the effects of curcumin combined with cisplatin on the expressions of Bcl-2 and caspase-3 in the lung cancer A549 cells. MethodsThe lung cancer A549 cells were divided into the control group, curcumin group, cisplatin group and combination group. Each group was treated with the corresponding drugs. The inhibition rate of cell proliferation was detected by MTT assay. The cell apoptosis rate and cell cycle were detected by flow cytometry. The expression levels of Bcl-2 mRNA and caspase-3 mRNA were detected by RT-PCR,and the protein expression levels of Bcl-2 and caspase-3 were detected by Western blot. Results:①Compared with the control group,the inhibition rates of cell proliferation were increased in the other groups after 24-,48- and 72-hour drug treatment,and the inhibition rate of the combination group was higher than that of the curcumin group or cisplatin group,with statistically significant differences(P<0.05). ②Compared with the control group,the cell apoptosis rates were increased in the other groups,and the apoptosis rate of the combination group was higher than that of the curcumin group or cisplatin group(P<0.05). The cell arrest in G2/M phase was observed in the curcumin group,and the cell arrest in S phase was found in the cisplatin group and combination group. ③Compared with the control group,the mRNA and protein expression levels of Bcl-2 in lung cancer A549 cells were decreased in the other groups. The expression levels in the combination group were lower than those in the cisplatin group,and the expression levels in the cisplatin group were lower than those in the curcumin group(P<0.05). ④Compared with the control group,the mRNA and protein expression levels of caspase-3 in lung cancer A549 cells were increased in the other groups. The expression levels in the combination group were higher than those in the cisplatin group,and the expression levels in the cisplatin group were higher than those in the curcumin group(P<0.05). Conclusion:Curcumin combined with cisplatin can inhibit the expression of Bcl-2 and promote the expression of caspase-3 in lung cancer A549 cells,and thus inhibit the cell proliferation and promote the cell apoptosis.
