Effects of Zuogui Drink on morphology of liver and kidney tissues and expression of apoptosis related proteins in D-galactose induced aging rats
Objective:To study the effects of Zuogui Drink on the morphology of liver and kidney tissues and expression of apoptosis related proteins in D-galactose induced subacute aging rats,and discuss its mechanism of anti-aging. Methods72 healthy SD rats(half male and half female)were randomly divided into the control group,model group,vitamin E group,Liuwei Dihuang Pill group and Zuogui Drink groups with high- and low-dose. Except the control group that treated with 0.9% NaCl solution by subcutaneous injection at the neck back,the other groups were treated with D-galactose by continuously subcutaneous injection at the neck back to establish the subacute aging model. During the modeling,the rats in the drug treatment groups were treated with vitamin E,Liuwei Dihuang Pill and Zuogui Drink with high- and low-dose by intragastric administration,respectively,and the control and model group were treated with distilled water at equal volume. After administration for continuous 8 weeks,the liver and kidney tissue of rats were collected and made into the pathological sections for microscopic observation after HE staining. The positive expression levels of apoptosis related proteins(Bax,Bcl-2 and Caspase-3)in the liver and kidney tissue were detected by immunohistochemistry,and the IOD values were analyzed and compared. Results:①Compared with the control group,there were obvious pathological injuries of liver and kidney tissue in the model group. Compared with the model group,Zuogui Drink could alleviate the pathological damage of liver and kidney tissue in the aging process induced by D-galactose in rats. ②Compared with the control group,the IOD values of Bax and Caspase-3 in the liver were significantly increased in the model group(P<0.01),and the IOD value of Bcl-2 was significantly decreased(P<0.01). Compared with the model group,the IOD values of Bax and Caspase-3 in the liver were significantly decreased in the Zuogui Drink group with high-dose(P<0.01),and the IOD value of Bcl-2 was significantly increased(P<0.01). Compared with the model group,the IOD values of Bax and Caspase-3 in the liver were significantly decreased in the Zuogui Drink group with low-dose(P<0.05),the IOD value of Bcl-2 was slightly increased,but with no statistically significant difference(P>0.05). ③Compared with the control group,the IOD values of Bax and Caspase-3 in the kidney were significantly increased in the model group(P<0.01),and the IOD value of Bcl-2 was significantly decreased(P<0.01). Compared with the model group,the IOD values of Bax and Caspase-3 in the kidney were obviously decreased in the Zuogui Drink group with high-dose(P<0.01),and the IOD value of Bcl-2 was obviously increased(P<0.01). Compared with the model group,the IOD value of Bax in the kidney was obviously decreased in the Zuogui Drink group with low-dose(P<0.05),but there were no statistically significant differences on the IOD values of Bcl-2 and Caspase-3(P>0.05). Conclusion:Zuogui Drink shows a certain protective effects on the liver and kidney tissue,and its mechanism may be associated with the expression of apoptosis related proteins in the liver and kidney tissue.
