Experimental study of the effects of Gegen Qinlian Pill on mouse model of rotavirus diarrhea with dampness-heat syndrome
Objective:To observe the effects of Gegen Qinlian Pill on mose model of rotavirus diarrhea with dampness-heat syndrome(RV-DH)and the mechanism on immune barrier dysfunction of intestinal mucosa. Methods80 BLAB/C mice were randomly divided into 8 groups. The mouse model of syndrome of dampness-heat in large intestine and mouse model of RV-DH with syndrome of dampness-heat in large intestine were established respectively. After successful modeling,except the model group,the other groups were treated with Gegen Qinlian Pill,montmorillonite powder(Smecta)and drug combination respectively. The intestinal mucosal shielding function in mice was detected by FITC-Dextran. The levels of SIgA,IL-1β,IL-2,IL-6,IL-8 and TNF-α were detected by ELISA. The pathology of large intestine in mice was observed by HE staining. Results:①The FITC-Dextran detection showed that,in mouse model of syndrome of dampness-heat in large intestine,the FD4 level was decreased in the Chinese medicine group,Smecta group and combination group compared with the model group,and the therapeutic efficacy of the combination group was the most significant. In mouse model of RV-DH with syndrome of dampness-heat in large intestine,the trends in the treatment groups after drug intervention were consistent with those in mouse model of syndrome of dampness-heat in large intestine. ②The ELISA detection indicated that,in mouse model of syndrome of dampness-heat in large intestine,the levels of SIgA,IL-1β,IL-2,IL-6,IL-8 and TNF-α in large intestine and lung tissue were decreased in the Chinese medicine group,Smecta group and combination group compared with the model group,and the efficacy of the combination group was the most significant. In mouse model of RV-DH with syndrome of dampness-heat in large intestine,the trends in the treatment groups after drug intervention were consistent with those in mouse model of syndrome of dampness-heat in large intestine. The intestinal mucosal barrier function in mice was improved in all drug treatment groups. ③The observation of HE staining suggested that the acute inflammatory reactions were alleviated in all drug treatment groups,and the number of infiltrating lymphocytes in submucosa was decreased and the intestinal mucosal damage was improved. The efficacy of the combination group was the most significant. Conclusion:Gegen Qinlian Pill shows certain effects of immune regulation. It can regulate lymphocytes and inhibit TNF-α in intestinal homogenate,which may be one of its mechanisms in the treatment of rotavirus diarrhea with dampness-heat syndrome.
