Protective effects of Xiaochaihu Decoction and Danggui Shaoyao Powder with single and combined application on immunological liver injury
Objective:To investigate the protective effects of Xiaochaihu Decoction and Danggui Shaoyao Powder with single and combined application on immunological liver injury. Methods Seventy-two KM mice were randomly divided into the blank group, model group, Xiaochaihu Decoction(XCHD,6.63 g/kg) group, Danggui Shaoyao Powder(DGSYP,11.31 g/kg)group, combination(Chaigui Decoction,17.94 g/kg)group and positive control group (bifendate 0.2 μg/g and diammonium glycyrrhizinate 0.3 μg/g). Except the blank group, the model of immunological liver injury was established in the other group by intravenous injection of concanavalin A(Con A)in mouse tail. Each group was treated with the corresponding intervention for 7 days. The levels of ALT, AST, ALB and TP were detected by semi-automatic biochemical analyzer. The pathological section was observed by HE staining. The activity of SOD and the contents of GSH and MDA in liver tissue were determined by colorimetry. The levels of TNF-α,IL-1 and IL-6 were detected by ELISA. The protein expressions of Bax, Bcl-2 and Caspase-3 were detected by Western blotting. The T lymphocyte subsets were detected by APAAP method. Results: Compared with the model group, XCHD could decrease the ALT and AST levels, reduce the IL-1 level, decrease the MDA content, and Chaigui Decoction significantly decrease the Bcl-2 protein expression. Compared with the model group, DGSYP could decrease the ALT level, reduce the serum levels of TNF-α,IL-1 and IL-6, and reduce the protein content of Bcl-2. Chaigui Decoction showed significantly protective effects. The effects on improving above indicators in the combination group were better than those in the XCHD group and DGSYP group,and Chaigui Decoction significantly improved the ratio of CD4,+,/CD8,+, in serum. Conclusion: Xiaochaihu Decoction and Danggui Shaoyao Powder show the protective effects on ConA induced immunological liver injury respectively. The combination of the two prescriptions shows better effects than the single application. The mechanism may be associated with promotion on anti-oxidation,anti-apoptosis and regulation of immune disorders.
