Study on anti-tumor effects of Compound Purple Ginseng polysaccharides on H22 tumor-bearing mice
Objective:To study the inhibitory effect and mechanism of Compound Purple Ginseng polysaccharides(CPGP)in hepatocarcinoma 22(H,22,)transplanted mice. Methods 60 male ICR mice were selected and H,22, tumor-bearing mice model was established except the blank group. Then the mice were randomly divided into the model group,cyclophosphamide(CTX,25 μg/g),CPGP groups with low-(70 μg/g),middle-(140 μg/g),high-(280 μg/g)dose,10 mice in each group. Each group were given the corresponding intervention,with a course of 10 days. The general condition of mice was observed. The tumor inhibition rate and organ indexes were calculated. The serum levels of TNF-α,IL-2,IFN-γ were determined by ELISA. The T-lymphocyte subsets in spleen were detected by flow cytometry. Results: The tumor inhibition rates of CTX group and CPGP groups with low-, middle- and high-dose on H,22, were 65.01%、40.19%、51.32%、57.84% respectively. Compared with the model group,the body weight and immune organ indexes were significantly increased in CPGP groups with low-, middle- and high-dose(P,<,0.05,P,<,0.01),the serum levels of TNF-α,IL-2,IFN-γ were increased in CPGP group with high-dose(P,<,0.05),and the ratio of CD4,+,/CD8,+, in splenic T-lymphocyte subsets was increased in CPGP groups with low-, middle- and high-dose(P,<,0.01).Compared with the model group,the body weight and immune organ indexes,the serum levels of TNF-α,IL-2,IFN-γ and the ratio of CD4,+,/CD8,+, in splenic T-lymphocyte subsets were significantly decreased in CTX group(P,<,0.05,P,<,0.01).Compared with CTX group,the body weight and immune organ indexes,the serum levels of TNF-α,IL-2,IFN-γ and the ratio of CD4,+,/CD8,+, in splenic T-lymphocyte subsets were significantly increased in CPGP-treated mice(P,<,0.05,P,<,0.01). Conclusion: CPGP shows significantly inhibitory effects on transplanted H,22, tumor in mice. Its mechanism may be associated with enhancing immune function.
