目的:探讨调心方对APP/PS1双转基因AD小鼠学习记忆能力和海马CA1区突触和神经元结构、突触数量的影响。 方法:将54只3月龄APP/PS1双转基因AD小鼠随机均分为模型组、调心方组(1.485 g·kg-1·d-1)和盐酸多奈哌齐组(0.083 mg·kg-1·d-1),以同月龄同性别下18只C57BL/6J野生小鼠作为正常组,各组采取相应干预措施。灌胃12周后,采用Morris水迷宫进行行为学检测,采用透射电镜观察小鼠海马CA1区突触和神经元结构、突触数量和神经元。 结果:①与正常组比较,模型组小鼠逃避潜伏期的时间明显延长(P<0.01),穿越平台的次数明显减少(P<0.05),在原平台象限停留的时间明显减少(P<0.01);海马CA1区神经元细胞核固缩,核膜皱缩增厚,核周出现空晕,部分临近胞膜处染色较深,异染色质,线粒体肿胀,粗面内质网扩张呈囊泡状,神经毡内突触数量明显减少(P<0.01),突触结构出现大片的轴浆空泡化。②与模型组比较,调心方组小鼠逃避潜伏期的时间明显缩短(P<0.01),穿越平台的次数明显增加(P<0.01),在原平台象限停留的时间明显增加(P<0.01);海马CA1区神经元细胞核圆形,核膜清晰完整,核仁明显,染色质细腻,线粒体嵴清晰可见,线粒体部分略微肿胀,粗面内质网和核糖体都较正常,突触结构较清晰,突触数量明显增加(P<0.01),细胞器病变有所减轻。 结论:调心方可改善APP/PS1双转基因AD小鼠学习记忆水平,改善神经元和突触结构的退变和突触数量的减少,对认知和突触具有保护作用。
Objective:To investigate the effects of Tiaoxin Recipe on the learning and memory ability of APP/PS1 transgenic AD mice and synapse and neuronal structure and synapse number in hippocampal CA1 region. MethodsFifty-four APP/PS1 double transgenic AD mice with three months old were randomly divided into the model group,Tiaoxin Recipe group and donepezil hydrochloride group,and 18 wild C57BL/6J mice with the same age and sex were taken as the normal group. Corresponding intervention were given in each group. After intragastric administration for 12 weeks,the behavioral test was performed by Morris water maze,and the synapse and neuronal structure and synapse number in hippocampal CA1 region were observed by transmission electron microscopy. Results:①Compared with the normal group,the escape latency of mice in the model group was significantly prolonged (P<0.01),the number of platform crossing was significantly reduced (P<0.05),the stay time in the original platform quadrant was significantly decreased(P<0.01). The neuronal nuclear in hippocampus CA1 region showed pyknosis,the nuclear membrane was shrunk and thickened and the perinuclear empty halo was found. Some regions near the cell membrane were stained deeply,and there were heterochromatin and swelling mitochondria. The rough surfaced endoplasmic reticulum showed expansion as vesicle shape,the number of synapses in the neuropil was decreased significantly(P<0.01),and large axoplasmic vacuoles were found in synaptic structures. ②Compared with the model group,the escape latency of mice in the Tiaoxin Recipe group was significantly shortened (P<0.01),the number of platform crossing was significantly increased (P<0.01),the stay time in the original platform quadrant was significantly increased(P<0.05). The neuronal nucleus in hippocampus CA1 region were round with clear and complete nuclear membrane and obvious nucleolus. The chromatin was fine and smooth,the mitochondrial cristae was clearly visible and parts of mitochondria were slightly swollen. The rough surfaced endoplasmic reticulum and ribosomes were normal,the synapse structure was clear and the number of synapses was significantly increased (P<0.01),and the organelles lesion were alleviated. Conclusion:Tiaoxin Recipe can improve the learning and memory ability of APP/PS1 transgenic AD mice,and improve the degeneration of neuron and synapse structure and reduction of synapses number,which shows protective effects on cognition and synapse.
