目的:观察红珊瑚对实验大鼠骨折愈合过程的影响。 方法:60只SPF级Wistar大鼠随机取12只为空白对照组,其余大鼠造成左前肢桡骨骨折模型,经X射线影像透视后确定大鼠桡骨骨折模型建立成功。将骨折模型大鼠随机分为模型组、阳性药物(伤科接骨片,150 μg/g)组、红珊瑚高剂量组(300 μg/g)、红珊瑚低剂量组(150 μg/g),每组12只。造模后第2 d开始灌胃药,每天1次。给药后第10 d、20 d、30 d分别对每组大鼠骨折部位进行X射线影像评估,骨折部位HE染色后观察组织病理改变,并用酶联免疫法(ELISA)测定血清中碱性磷酸酶(ALP)活性及钙(Ca)、磷(P)含量。 结果:X射线影像显示,给药第10 d时红珊瑚高剂量组大鼠骨折端边缘线变模糊;20 d时骨折断端骨痂增多,密度增加,边缘接近消失;30 d时骨痂填满骨折缺损,完全愈合。骨折部位HE染色可以看出红珊瑚高剂量组大鼠骨折处纤维组织多,骨痂形成速度较其他组更快,红珊瑚高剂量组的骨折愈合周期较其他组更短。红珊瑚高剂量组血清中Ca、P水平逐渐升高,并明显高于模型组,差异均有统计学意义(P<0.05)。ALP含量有先升后降趋势,较其他组先出现且明显,差异有统计学意义(P<0.05)。 结论:红珊瑚能够调节大鼠血清ALP和Ca、P水平,加速钙盐沉积,促进大鼠骨折部位纤维组织增多,加速骨痂形成,缩短骨折愈合周期,对骨折的愈合和修复有促进作用。
Objective:To observe the effects of red coral (Corallium rubrum) on the healing of experimental rat fracture. Methods 12 rats were randomly selected as the blank group from sixty Wistar rats with SPF grade,and the radius fracture model on left forelimb was established in the other rats and verified by X-ray fluoroscopy. The successful modeling rats were randomly divided into the model group,positive drug group(150 μg/g),red coral groups with low-dose(150 μg/g) and high-dose(300 μg/g),12 rats in each group. From the second day after modeling,the drugs were given to the corresponding group by intragastric administration,once a day. After administration for 10,0,30 days,the fracture sites of rats in each group were evaluated by X-ray fluoroscopy and the histopathological changes were observed after HE staining. The serum contents of alkaline phosphatase (ALP), calcium (Ca) and phosphorus (P) were detected by enzyme linked immunosorbent assay(ELISA). Results: The X-ray image showed that the edge line of fracture became blurred in the red coral group with high-dose after administration for 10 days;when administration for 20 days,the callus increased in the broken end of fracture and the density increased,the edge almost disappeared;when administration for 30 days,the callus filled with the fracture defect and complete healing was found. The HE staining of the fracture site showed that more fibrous tissue was found in the red coral group with high-dose,the callus formation rate was faster than that in the other groups,and the fracture healing time was shorter than that in the other groups. The serum levels of Ca and P were gradually increased in the red coral group with high-dose,and significantly higher than those in the model group,with statistical differences(P< 0.05). The content of ALP was increased first and then decreased in the red coral group with high-dose,and this change was more earlier and significant than that in the other groups, with statistical difference(P< 0.05). Conclusion: The red coral can regulate the serum levels of ALP,Ca and P,accelerate calcium salt deposition,promote the increase of fibrous tissue in the fracture site of rats,accelerate the callus formation,shorten the healing period of fracture,which shows promotion effects on the healing and repair of fracture.
