Isolation and purification of Asperosaponin Ⅵ from Dipsacus asper Wal. by high-speed counter-current chromatography
Objective:To establish the high-speed counter-current chromatography (HSCCC) method for isolation and purification of Asperosaponin Ⅵ from Dipsacus asper Wal.. Methods Dried root of Dipsacus asper Wal. is taken as raw material. The ethanol extracts are first treated with AB-8 macroporous resin to enrich target substances. Then Asperosaponin Ⅵ is isolated and purified by semi preparative HSCCC. A mixture of ethyl acetate-n-Butanol-water (3∶1∶4,v/v) is used as the two phase solvent system,which the light phase is the stationary phase and the heavy phase is the mobile phase with flow rate of 3.0 ml/min. The speed of apparatus is 800 r/min. The evaporative light scattering detector (ELSD) was used as the detector. Results: After enrichment by macroporous adsorption resin and isolation by HSCCC,75 mg Asperosaponin Ⅵ is obtained from 200 mg extract of Dipsacus asper Wal. . The purity is 98.5 % by HPLC detection. Conclusion: This method is quick and simple with good repeatability,which provides reference for mass production of Asperosaponin Ⅵ.
