Effects of Saikosaponin d on mitochondrial membrane potential and apoptosis in human neuroblastoma SH-SY5Y cells
Objective:To investigate the effects of Saikosaponin d (SSd) on mitochondrial transmembrane potential (△ψm) and apoptosis in human neuroblastoma cells (SH-SY5Y). MethodsSH-SY5Y cells were cultured in high glucose DMEM; and SSd was dissolved in DMSO. SH-SY5Y cells were co-cultured with 0~10 μmol/L SSd for 48h. the △ψm of SH-SY5Y cells were analyzed with flow cytometry by Rh123 staining. At the same time, apoptosis levels of cells were analyzed by acridine orange (AO)/ethidium bromide (EB) staining and TUNEL Kit. Results:The fluorescence intensity of Rh123 was reduced significantly in SH-SY5Y cells after treatment with SSd for 48 h. It also showed that 8-10 μmol/L of SSd induced the increase of apoptotic index of SH-SY5Y cells by TUNEL staining (P<0.01). Conclusion: SSd can promote the apoptosis of SH-SY5Y cells by reducing △ψm.
