Objective:To investigate the effects and mechanisms of “Qufeng Zhitong Ointments” on adjuvant arthritis (AA) in rats. MethodsArthritis severity was evaluated by body weight loss, paw swelling, arthritic score, and histological changes. In addition, the serum levels of TNF-α, IL-1, SOD, MDA, GSH and NO were detected using ELISA method. Another portion of blood was used for blood routine examination . The expressions of MMP-10 and VEGF in rat cartliage tissue were detected using immunohistocheical analysis. Moreover, the cell culture supernatants of celiac macrophage cells isolated from AA rats with or without treatment of the medicine were added to mice thymic cells and rats splenic cells, and the proliferation of mice thymic cells and rats splenic cells were observed by MTT assay.  Results:Compared with the model group “Qufeng Zhitong ointments” significantly ameliorated paw swelling and arthritis score, down-regulated the levels of MDA,IL-1,TNF-α,NO and up-regulated the SOD and GSH levels in serum(P<0.05,P<0.01). It also inhibited the proliferation of splenic cells and mice thymic cells, effectivelly releasedjoint hyperplasia, and down-regulated MMP-10 and VEGF expression in crtliage tissue(P<0.05,P<0.01).  Conclusion:The results showed that Qufengzhitong ointments has good effect on adjuvant arthritis, the mechanism may be related to recucing oxidative damage, inhibitng MMP-10 and VEGF expression.
