Optimization of extraction technique of compound Shi-Lin-Tong capsule based on analytic hierarchy process of multi-index comprehensive evaluation
Objective:To optimize the extraction techniques of compound Shi-Lin-Tong capsule and compare it with its original extraction technique. Methods The orthogonal test was used by two evaluating methods (the first method was to determine the contents of chlorogenic acid and schaftoside and the extraction rate; the second method was to determine the HPLC characteristic fingerprint peak area and extraction rate) to investigate the influence of three factors on extraction factors(solvent doses, extraction time and alcohol concentration; and the single factor test was used to investigate the effect of extraction frequency on the transfer rate of active ingredients. The analytic hierarchy technique was used to determine weight coefficients of each index and to evaluate the results of the test. The transfer rates of chlorogenic acid and schaftoside as well as the difference of two extraction techniques were compared. Results: The outcomes obtained from the two evaluating methods were consistent. The optimized extraction technique of Shi-Lin-Tong capsule was as follows:the medicinal materials were extracted with ten times of water at the first extraction and six times of water at the second extraction, one hour for each time; and the concentrated solution was added to anhydrous ethanol until the ethanol concentration reaching 80% for ethanol precipitation. The transfer rates of chlorogenic acid and schaftoside of the optimized extraction technique was significantly higher than the original extraction technique. Conclusion: The optimized extraction technique is feasible, stable, and repeatable, and facilitates to extract the effective components of compound Shi-Lin-Tong capsule and suits for industrial production.
