The puerarin impact on the expression of ABCG2 in human renal proximal tubule epithelial cells
Objective:To explore the influence of Puerarin(Pur) on expression of gene andprotein of ATP-binding cassette superfamily G member2(ABCG2) in human renal proximal tubular epithelial cells(HK2),and to clarify Pur the possible mechanism for the treatment of gout. MethodsHK2 cells were cultured in vitro,and were divided into control group and 25, 50 , 100, 200 mg/L Pur dose groups. CCK-8 method was used to detect Pur effects on HK2 cells; ATP content was detected, HK2 cells enzymatic reaction condition was observed; Fluorescence quantitative PCR and Western blotting methods were used for determination HK2 ABCG2 gene protein expression in the control group and the different dose groups. Results:Pur has promote proliferation effect on HK2 cells(P<0.05). The best group of ATP activity was 100 mg/L in dose group; detection by Real-time PCR and Western blotting methods showed that HK2 cells ABCG2 gene and protein expression in dose group was significantly higher than that of the control group,of which 100 mg/L of dose group was most obvious(P<0.01). Conclusion:Pur could increase HK2 cells the expression of ABCG2,which indicated that this maybe one of the important molecular mechanism for regulation gout patients blood uric acid level.
