Influence of the“Dingxiang Jiangqi Decoction” on IL-23/IL-17 inflammation axis in rats with reflux esophagitis
Objective:To observe influence of the “Dingxiang Jiangqi Decoction” on IL-23/IL-17 inflammation axis in rats with reflux esophagitis. Methods40 Spregue-Dawley(SD) rats were randomly divided into treatment group,control group,model group and normal group, 10 in each group. Reflux esophagitis models were induced by cardiomyotomy and pylorus semiligation,and given the corresponding intervention measures. Pathological changes of esophagus tissues were observed by light microscope in each group, and the expression of IL-23,I L-17 mRNA in esophageal tissues of rats were detected by real-time RT-PCR, IL-17 protein content in esophageal tissues was measured by ELISA. Results:Compared with the normal group, the esophageal mucosa in each group had different degrees of inflammation,but the Dingxiang Jiangqi Decoction group was mild than the western medicine group(P<0.05).The expressions of IL-23 and IL-17 mRNA and IL-17 protein in the esoph ageal tissues of model group were significantly higher than the normal group(P<0.05). Compared with model group, the expressions of IL-23 and IL-17 mRNA and IL-17 protein in the esoph ageal tissues of Chinese medicine group were lower(P<0.05), and the expression of IL-23mRNA in western medicine was lower(P<0.05). Compared with western medicine group, IL-17 protein decreased and IL-17 mRNA expression decreased(P<0.05). There was no significant difference of IL-17 protein expression between the Dingxiang Jiangqi Decoction group and the normal group(P>0.05). There was no significant difference in IL-23mRNA expression in Dingxiang Jiangqi Decoction group,control group and normal group(P>0.05); while there was no significant difference between the Dingxiang Jiangqi Decoction group and the western medicine group(P>0.05). Conclusion:“Dingxiang Jiangqi Decoction” can effectively improve the esophageal mucosal inflammation of reflux esophagitis rats,the regulation of the IL-23/IL-17 inflammation axis may be one of the mechanisms of the “Dingxiang Jiangqi Decoction”.
