摘要

目的：观察消痰散结方对结肠癌细胞株细胞周期分布的影响,并探讨其相关机制。方法：消痰散结方浸膏冻干粉末溶解在细胞培养液中,制备消痰散结方保存液。体外以不同浓度消痰散结方(0.24 g/L、0.47 g/L、0.94 g/L、1.88 g/L)作用于HCT116细胞72 h后,流式细胞仪检测细胞周期分布；Western blotting法检测药物干预后细胞周期蛋白(cyclinA2、cyclinB1)和细胞周期蛋白依赖性激酶蛋白(cyclin dependent kinase,CDK2、CDK1)的表达变化。结果：消痰散结方作用HCT116细胞72 h后,0.47 g/L、0.94 g/L组S期细胞比例增高,0.47 g/L、0.94 g/L、1.88 g/L组G,2,期细胞比例增高,与对照组比较差异有统计学意义(P,<,0.01)。消痰散结方作用72 h后,0.24 g/L、0.47 g/L、0.94 g/L、1.88 g/L组CDK2、cyclinA2、CDK1表达显著降低,0.47 g/L、0.94 g/L、1.88 g/L组cyclinB1表达显著降低,与对照组比较差异有统计学意义(P,<,0.01),且效应呈浓度依赖性。结论：消痰散结方对HCT116细胞增殖的抑制作用是通过下调cyclinA2、cyclinB1和CDK2、CDK1的表达,使CDK2/cyclinA2、CDK1/cyclinB1复合体活性减弱,阻滞细胞周期于S和G,2,期实现的。

Abstract

Objective:To observe the effect of “Xiaotan Sanjie Recipe” on the cell cycle distribution of colorectal cancer cell line,and to study the possible mechanisms. MethodsThe original “Xiaotan Sanjie Recipe” aqueous solution was lyophilized, weighted and dissolved in cell culture and stored. HCT116 cells were treated with different concentrations of “Xiaotan Sanjie Recipe” for 72h,cell cycle distribution was detected by flow cytometry,the expression of proteins(cyclinA2,cyclinB1,CDK2 and CDK1)were detected by Western Blotting. Results:After being treated by “Xiaotan Sanjie Recipe” for 72 h,the proportion of the cells in S phase was increased in 0.47 and 0.94 g/L group, the difference was significant compared with the controlled group(P,<,0.01); the proportion of the cells in G,2, phase were increased in 0.47,0.94 and 1.88 g/L group,the difference was significant compared with the controlled group(P,<,0.01). After being treated by “Xiaotan Sanjie Recipe” for 72 h,the expressions of cyclinA2, CDK2 and CDK1 proteins were significantly decreased in 0.24,0.47,0.94 and 1.88 g/L group,the expression of cyclinB1 was significantly decreased in 0.47,0.94 and 1.88 g/L group,the difference was significant compared with the controlled group(P,<,0.01),the effect was concentration-dependent. Conclusion:“Xiaotan Sanjie Recipe” has the ability to induce the S phase and G,2, phase arrest of HCT116 cell line,its mechanism might be down-regulating the expression of cyclinA2,cyclinB1,CDK2 and CDK1 proteins.
