摘要

目的：探讨中药复方肠胃清(CWQ)逆转人结肠癌HCT116/L-OHP细胞耐药的分子机制。方法：提取肠胃清含药血清,设置不同浓度,采用MTT法确定肠胃清的最大无毒剂量和逆转指数(RI)；采用彗星实验法检测肠胃清联合奥沙利铂(L-OHP)对耐药细胞DNA损伤的影响,流式细胞术观察肠胃清联合奥沙利铂(L-OHP)对耐药细胞凋亡的影响。结果：肠胃清含药血清可以逆转人结肠癌HCT116/L-OHP耐药细胞对L-OHP的耐药性,逆转指数(RI)为 2.63倍。肠胃清联合L-OHP能促进耐药细胞发生DNA损伤(P<0.001)及凋亡(P<0.01)。结论：肠胃清能够逆转人结肠癌铂类化疗耐药,其机制与促进肿瘤细胞DNA损伤、诱导凋亡有关。

Abstract

Objective:To investigate the molecular mechanisms of resistance of traditional Chinese medicine Changwei Qing(CWQ)reversal of human colon cancer oxaliplatin. MethodsExtract CWQ containing serum,using MTT assay to detect CWQ reversal index(RI),using the comet assay to detect cell DNA damaged and detect cell apopotosis by flow cytometr in resistant cells. Results:CWQ drug serum containing reversing HCT116/L-OHP resistant cells to L-OHP resistance reversal index(RI)was 2.63 times. The CWQ L-OHP group of resistant cells could promote DNA damage(P<0.001)and apoptosis(P<0.01). Conclusion:CWQ can reversal drug resistance of human colon cancer to Platinum-based Chemotherapy,and it's mechanism may has relationship with promotion DNA damage and apoptosis occurred in cancer cells.
