摘要

目的：观察消痰散结方对裸鼠人胃癌细胞SGC-7901皮下移植瘤Runt相关转录因子3(RUNX3)、核转录因子κB p65(NF-κB p65)蛋白表达的影响,探讨消痰散结方的抗胃癌作用及机制。方法：建立裸鼠人胃癌细胞SGC-7901皮下移植瘤模型,随机分为中药组(消痰散结方)、化疗组(卡培他滨)及对照组(0.9%NaCl溶液),每组10只,各组给予相应干预共32天。观察各组裸鼠体质量、瘤体积、瘤重,并采用Western blotting法检测各组裸鼠皮下移植瘤组织中RUNX3、NF-κB p65蛋白表达水平。结果：中药组及化疗组的瘤体积及瘤重值均低于对照组(P<0.05)；与中药组比较,化疗组的瘤体积及瘤重值均下降,差异有统计学意义(P<0.05)。中药组和化疗组的抑瘤率分别为44.91%和83.23%。中药组和对照组体质量差异无统计学意义(P>0.05)；化疗组体质量低于对照组及中药组(P<0.05)。中药组和化疗组的肿瘤组织RUNX3蛋白表达水平高于对照组、NF-κB p65蛋白表达水平低于对照组(P<0.05)。结论：消痰散结方具有较好的抗胃癌疗效,虽然在抑瘤率方面不及卡培他滨,但没有引起体质量下降的副作用；上调RUNX3蛋白并下调NF-κB p65 蛋白表达水平是其可能的作用机制之一。

Abstract

Objective:To observe the effect of “Xiaotan Sanjie Decoction” on the RUNX3 and NF-κB p65 protein expression in Human gastric cancer cell SGC-7901-bearing nude mice,and discuss the anti-gastric cancer effect and mechanism of “Xiaotan Sanjie Decoction”. MethodsHuman gastric cancer cell SGC-7901-bearing nude mice models were made. The mice were divided into TCM group (“Xiaotan Sanjie Decoction”),chemotherapy group (capecitabine) and control group (0.9%NaCl saline) randomly, 10 in each group, with 32 d intervention respectively in each group. The tumor volume,tumor weight, body weight, RUNX3 and NF-κB p65 protein expression in tumor were observed the the method of Western blotting. Results:The tumor volume and weight of TCM group and chemotherapy group were lower than those of control group(P<0.05); compared with TCM group, the tumor volume and weight in chemotherapy group decreased, and the difference was statistically(P<0.05); The anti-tumor rate of TCM group and chemotherapy group were 44.91% and 83.23% respectively. There was no statistical significance in weight between TCM group and control group(P>0.05). The body weight in chemotherapy group decreased compared with that in control group and TCM group(P<0.05). The RUNX3 protein expression in TCM group and chemotherapy group was higher than that in control group,while the NF-κB p65 expression was lower than that of control group(P<0.05). Conclusion:“Xiaotan Sanjie Decoction” has the good anti-gastric cancer effect. Although the anti-cancer rate of “Xiaotan Sanjie Decoction” was not as good as capecitabine,it had not bodyweight decreasing side effect. Increasing the RUNX3 expression and decreasing the NF-κB P65 protein expression in tumor tissues may account for the anti-gastric cancer mechanism of “Xiaotan Sanjie Decoction”.
