Effects of solanine in vitro induction SW480 cell apoptosis and related factors of human with colon cancer
Objective:To observe the effects of solanine on SW480 cell apoptosis of human with colon cancer and the related factors in ERK/MEK, PI3K/AKT signal pathway. MethodsThe SW480 colon cancer cell was divided into 5 groups after the cultivation, blank control group, culture solution control group and solanine 4,8, 16 mmol/L groups, each group administered with intervention respectively. The proliferation function of solanine on colon cancer cell line was tested with MTT method, the effects of solanine on colon cancer cell apoptosis was tested with FCM, the expression of ERK/MEKand PI3K/AKT in SW480 cell that treated with solanine was detected with RT-PCR and Western blotting method. Results:MTT growth curve showed that solanine could inhabite the proliferation of colon cancer SW480 cell line. Flow Cytometry showed that solanine could promote the SW480 cell apoptosis. RT-PCR showed that solanine could inhabite the expression of ERK1/2 mRNA. Western blotting showed that solanine could decrease the protein expression in the MEK/ERK and PI3K/AKT signal pathway. Conclusion:Solanine could inhabite the proliferation of colon cancer, and the cell apoptosis induction was realized may be though MEK/ERK and PI3K/AKT signal pathway.
