目的:观察黄芪桂枝五物汤抗心肌细胞缺氧/复氧损伤的影响,探讨其对心肌缺血再灌注损伤的保护机制 方法:取体外培养的大鼠乳鼠心肌细胞建立缺氧/复氧心肌细胞损伤模型,分为6组:正常组,模型组,阳性对照组,黄芪桂枝五物汤高、中、低剂量组。正常组和模型组灌胃等量生理盐水;阳性对照组灌胃等量复方丹参滴丸溶液(0.075 g·kg-1·d-1);黄芪桂枝五物汤组灌胃等量高(9.0 g·kg-1·d-1)、中(4.5 g·kg-1·d-1)、低(2.25 g·kg-1·d-1)剂量黄芪桂枝五物汤煎剂;灌胃3天后腹主动脉取血,制备含药血清。使用含有不同浓度药物的低氧预处理1h的培养基,对各实验组建立缺氧复氧/损伤模型。观察黄芪桂枝五物汤对细胞缺氧/复氧后CK活性、NO含量、SOD活性、MDA 含量的影响。用Real-time PCR检测Bcl-2、Bax mRNA表达,Western blot检测Bcl-2、Bax蛋白表达 结果:黄芪桂枝五物汤能够降低心肌细胞缺氧/复氧损伤后CK活性、NO含量、MDA含量,升高SOD活性(P<0.05);减少Bax mRNA和蛋白表达(P<0.05)有增加Bcl-2蛋白表达的趋势(P>0.05) 结论:黄芪桂枝五物汤对心肌缺血再灌注损伤具有一定的保护作用。
Objective:To explore the effects of “Huangqi Guizhi Wuwu Decoction”(HGWD)on myocardial cell in ischemia reperfusion injury to study its protective function Methods:An anoxia/reoxygenation model of myocardial cells in Wistar rats was established.The experiment was divided into 6 groups:normal group, model group, control group,HGWD of high-, medium-and low-dose groups. Normal group and model group were given saline by gavage; control group was given same amount of Compound Danshen dripping pill solution by gavage(0.075 g·kg-1·d-1); HGWD groups were respectively given equivalent amount of high(9.0 g·kg-1·d-1),medium (4.5 g·kg-1·d-1) and low(2.25 g·kg-1·d-1) dose of HGWD; 3 days after intragastric administration,draw blood from abdominal aorta to prepare drug-containing serum. The use of drugs containing different concentrations of hypoxic preconditioning 1h medium, hypoxia reoxygenation injury model was set up for each experimental group. The CK activity, NO content, SOD activity and MDA content were observed; Realtime PCR was used to detect the expressions of Bcl-2 and Bax mRNA; Western blot was adopted to detect the expressions of Bcl-2 and Bax protein Results:“Huangqi Guizhi Wuwu Decoction” could reduce CK activity, NO and MDA contents, and increase SOD activity in myocardial cells with hypoxia reoxygenation injury; it could also reduce theexpressions of Baxprotein and mRNA, and increase theexpressions of Bcl-2 mRNA and protein Conclusion:“Huangqi Guizhi Wuwu Decoction” exert protective effects on myocardial cells with ischemia reperfusion injury.
