Effect of “Yiqi Qinghua Decoction” on intestinal flora in non-alcoholic fatty liver disease rats
Objective:To study the effect of “Yiqi Qinghua Decoction” on intestinal flora in HFD-induced non-alcoholic fatty liver disease rats model Methods:30 adult clean healthy Wistar rats was selected and randomly divided into blank group,model group and treatment group, 10 in each group. Rats in model group and treatment group were fed with high fat diet to prepare for non-alcoholic fatty liver disease rats model, the treatment group were treated with “Yiqi Qinghua Decoction” at the same time in the first day while model group were treated with 0.9% NaCl solution with the same quantity. Rats in blank group were treated with general diet and 0.9% NaCl solution with the same quantity. The course of each group was 8 weeks. At the end of the experiment,liver function and LPS of rats were tested, liver were collected for observing pathological changing with method of HE staining. Intestinal flora were determined with the method of ERIC-PCR fingerprint,and a cluster analysis on the fingerprint was performed. Result ① The serum ALT,AST, TG, TC and LPS levels in model group were significantly increased than that in the blank group(P<0.05), and the level of ALT, AST and LPS in the treatment group were significantly lower than that in the model group(P<0.05). ② There were a little fatty infiltration in the liver cells of the treatment group and inflammatory cells and necrosis of hepatic cells were significantly reduced in the treatment group than that in the model group. ③ The composition and distribution of bands of ERIC-PCR fingerprint in the treatment group was more and wider than that in the model group which showed that the structure of intestinal flora of the rats in the treatment group was partially recovered under the intervention of “Yiqi Qinghua Decoction”. ④ It was showed that the similarity of intestinal flora in the blank group and treatment group was greater than that of the blank and model groups and of the model and treatment groups. The data above showed that “Yiqi Qinghua Decoction” can correct the disorder of intestinal flora of non-alcoholic fatty liver disease rats. The analysis of intestinal flora diversity of rats showed that the diversity of blank group and treatment group were higher than that of model group, and it showed that “Yiqi Qinghua Decoction” can increase the species of intestinal flora of non-alcoholic fatty liver disease rats and then to improve the disorder of intestinal flora of non-alcoholic fatty liver disease rats Conclusion:“Yiqi Qinghua Decoction” can effectively alleviate injury of liver cells and improve steatosis on non-alcoholic fatty liver disease rats,and also can the disorder of intestinal flora of non-alcoholic fatty liver disease rats.
