摘要

目的：观察牛蒡子苷元(arctigenin,ARC-G)对关节软骨细胞增殖及Ⅱ型胶原表达的影响方法：采用酶消化法体外分离关节软骨细胞,取第1代软骨细胞分别接种于6孔培养板和96孔培养板中,实验分为空白组、牛蒡子苷元低浓度组(低浓度组)和牛蒡子苷元高浓度组(高浓度组)。各组分别给予相应的干预措施,48 h后,以SABC免疫组织化学染色法观察软骨细胞Ⅱ型胶原的表达情况,以MTT法检测细胞的增殖情况,以酶联免疫吸附法(ELISA)检测细胞上清Ⅱ型胶原的含量结果：①与空白组比较,低浓度组和高浓度组在软骨细胞数量和阳性染色强度上均有所增加,且高浓度组优于低浓度组；②与空白组比较,低浓度组和高浓度组软骨细胞OD值及Ⅱ型胶原含量均明显提高(P<0.05),且高浓度组优于低浓度组(P<0.05) 结论：牛蒡子苷元可明显促进体外培养关节软骨细胞的增殖和Ⅱ型胶原的表达,较好地维持软骨细胞表型,从而可有效用于骨关节炎的治疗。

Abstract

Objective:To observe effects of arctigenin on expression of joint chondrocytes proliferation and Ⅱ Collagen Methods:Articlular chondrocytes were isolated by enzyme digestion method and chondrocyteⅠwere inoculated into 6 well culture plate and 96 well culture plate. The test was divided into blank group, low and high Concentration arctigenin groups. After the intervention measures for 48 hours in each group, the expression of chondrocytesⅡ Collagen were observed by SABC method and chondrocyte proliferation was tested by MTT and content of Ⅱ Collagen was tested by ELISA Results:① Compared with the blank group, the quantity of chondrocytes and the positive staining potency of the low and high Concentration arctigenin groups were significantly increased, and the increasing range of high Concentration arctigenin group was higher than the low Concentration arctigenin group. ② Compared with the blank group,OD of Concentration and contents of Ⅱ Collagen in low and high Concentration arctigenin groups were significantly increased (P<0.05), and the increasing range of high Concentration arctigenin group was higher than the low Concentration arctigenin group (P<0.05) Conclusion:Arctigenin can significantly up regulate the expression of articular chondrocytes proliferation and Ⅱ Collagen and can maintain Concentration phaenotype which can be used in the treatment of osteoarthritis effectively.
