Analgesic effect of Toddalia Asiatica’s extract and its peripheral analgesic mechanism
Objective:To study the analgesic effect of Toddalia Asiatica, and to provide reference for the research and development of analgesic traditional Chinese medicine Methods:Analgesic effect of Toddalia Asiatica’s extract was investigated b Methods: of acetic acid writhing and hot plate, and the pain was induced by chemical and heat stimulation. ELISA kit was used to detect the regulation of beta-endorphin(β-EP), 5-hydroxytryptamine(5-HT), prostaglandin2(PGE,2,), nitric oxide(NO) in the serum of the rats; western blot was used to detect β-EP, PGE,2, and Opioid receptors μ in brain tissue of rats Results:The number of rats writhing induced by acetic acid was decreased by the Toddalia Asiatica’s extract, and the ethanol extract was less than the water extract (P,<,0.01). The pain threshold was remarkably improved by the Toddalia Asiatica’s extract, and there was no significantly difference between ethanol extract and water extract(P,>,0.05). The extract could increase the content of β-EP as well as reduce the content of PGE,2, and NO in the serum, and the ethanol extract has more remarkably effect(P,<,0.05); The extract also can increase the content of β-EP as well as reduce the expression of PGE,2, in brain tissue(P,<,0.05) Conclusion:Toddalia Asiatica has analgesic effect, and ethanol extract was better than water extract, high dose group was better than low dose group. The analgesic mechanism maybe has the relationship with increasing β-EP, decreasing the content of PGE,2, and NO, and the up-regulation of the expression of β-EP and the down-regulation of the expression of PGE,2,.
