Effect of “Jiawei Sangen Decoction” on function of osteoclast and expressions of IL-1β and TNF-α in osteoblasts-osteoclasts co-culture system
Objective:To detect the changes of tartrate-resistant acid phosphatase (TRACP), IL-1β and TNF-α in osteoblasts-osteoclasts co-culture system and to discuss its mechanism Methods:The osteoblasts-osteoclasts co-culture system model was established and SD rats were randomized into blank contol group, “Jiawei Sangen Decoction” groups. Six pores were set up in each group. The expressions of TRACP, IL-1β and TNF-α were detected by ELISA Results:The osteoblasts and osteoclasts were cultured and osteoblasts-osteoclasts co-culture system was set up. The TRACP in the medium and high dose “Jiawei Sangen Decoction” group were lower than that of the control group (P<0.05). Compared with the blank group, the IL-1β level in the medium and high dose “Jiawei Sangen Decoction” groups were reduced, and the TNF-α in the high dose “Jiawei Sangen Decoction” group was reduced significantly (P<0.05) Conclusion:“Jiawei Sangen Decoction” can inhibit the function of osteoclasts in osteoblasts-osteoclasts co-culture system, its mechanism may be associated with inhibiting the secretion of IL-1β and TNF-α.
