Effect of “Jiangqian I” on brain apoptosis of the offspring of rats exposed to low level lead exposure
Objective:To observe the intervention effect of “Jiangqian I” on brain apoptosis and mechanism study on improvment of neurobehavior Methods:Adult Wistar rats mated by female:male=2∶1 ratio were randomly divided into normal group and the experimental group which were given distilled water or 0.2% lead acetate solution respectively from the 1st day after pregnancy until the 21 days when the offspring began to be weaned. Weaning offspring is used for the experiment. Experimental rats were randomly divided into lead-exposed model group, EDTA group, the low dose “Jiangqian I” group, the middle dose “Jiangqian I” group and the high dose “Jiangqian I” group. The normal group used as blank group. Their blood and brain of lead was determined by atomic absorption spectrophotometry. Effects of learning and memory capacity of offspring after the medication were observed with Y-maze and apoptotic cells condition was detected with SP immunohistochemical method Results:After treatment, the blood lead levels of lead-exposed model group was significantly decreased after detachment of Lead Pollution (P<0.001). After treatment, the brain lead levels of EDTA group and the low dose “Jiangqian I” group were significantly lower than those of the model group (P<0.05). There are no significant difference between the brain lead levels of “Jiangqian I” group and those of EDTA (P>0.05); The rate of memory maintenance of the low and high dose “Jiangqian I” group were significantly higher than those of the model group (P<0.05, P<0.001). The rate of memory maintenance of the low dose “Jiangqian I” group were significantly higher than those of the EDTA group (P<0.01). The rate of memory maintenance of the low dose “Jiangqian I” group were significantly higher than those of the middle group (P<0.05); the apoptosis index of each treatment group were significantly lower than those of the model group (P<0.05, P<0.01) Conclusion:“Jiangqian I” can reduce blood and brain lead of lead exposed offspring, amend the impairment of brain tissue of lead exposed offspring in different degrees.
