Combination of Chinese herbal compound recipe and Tamoxifen on cell cycle and the expressions of CyclinD1, Bcl-2 and Bax protein of MCF-7 in breast cancer
Objective:To observe Chinese herbal compound recipe and Tamoxifen on cell cycle and the expressions of CyclinD1, Bcl-2 and Bax protein of MCF-7 in breast cancer Methods:Mice were randomly divided into the model group, Tamoxifen (TAM) group, Ru combined group (Ru yanning Recipe + TAM) and Yi combined group (Yi qixiaoji Recipe + TAM) after modeling successfully. All the mice were sacrificed through pull off neck after given drug for 21 days, tumor were taken completely, then cut for different specimens. The distribution of cell cycle was detected by Flow cytometry; the expression of Bcl-2, Bax and CyclinD1 proteins was detected by Western blot method Results:In each treatment group, the proportion of cells in G,0,/G,1, phase was increased, S phase was sharply reduced compared with the model group (P,<,0.01), but there was no statistical difference between Ru combined group and Yi combined group about cell ratio in the G,0,/G,1, phase and S phase (P,>,0.05); The expression of Bcl-2 and CyclinD1 protein was down-regulated, Bax was up-regulated in the three treatment groups, the ratio of Bcl-2/Bax was significantly decreased, the difference was significant compared with the model group (P,<,0.01), there was also significant difference between two combined groups and TAM group (P,<,0.01), but there was no significant difference between Ru combined group and Yi combined group (P,>,0.05) Conclusion:Chinese herbal compound formula has function of cell proliferation inhibition, the possible mechanism is related to blockcell cycle at G,0,/G,1, phase, down-regulate expression of CyclinD1, Bcl-2 protein and up-regulate the expression of Bax protein. There is synergy between Chinese herbal compound formula and TAM.
