Effect of laminarin on survival and apoptosis of human hepatocarcinoma cell of Bel-7402
Objective:To observe the effect of laminarin on survival and apoptosis of human hepatocarcinoma cell of Bel-7402.Methods① Cultured human hepatocarcinoma cell of Bel-7402 in vitro, after added with the 5,0, 20,0, 40 g/L laminarin, cultured for 48 h. Without adding laminarin as the control group, we observed the changes of cells by inverted microscope and detected the cell viability by MTT assay. ② Cultured humanhepatocarcinoma cell of Bel-7402 in vitro, three groups:The control group, laminarin group and laminarin+ medicine group were treated with 10,0 and 30 g/L respectively. The cell cycle and apoptosis rate were analyzed by flow cytometer. Results:① After bel-7402 cells were cultured using 5,0, 20,0, 40 g/L laminarin for 48 h, with the increasement of laminarin concentration, bel-7402 cells were shrinked and changes. The cell survival rate was significantly decreased. ② After 48h, the bel-7402 cells apoptosis rate was (14.8 ±1.7)% and death rate was (19.8±2.3), with significant difference from the control group (P<0.05). Conclusion:Laminarin can inhibit survival rate, and promote the apoptosis and necrocytosis of bel-7402 cells.
