目的:观察银杏内酯B药物洗脱支架在体内外模型中的抗血栓作用。 方法:①体外抗凝血试验检测:抽取健康成人全血,迅速置于含有2 ml枸橼酸钠抗凝剂的抗凝管中,然后倒入带有PLA-银杏内酯B的316L不锈钢槽与不带有PLA-银杏内酯B的不锈钢槽底部作用2 h,检测凝血活酶时间(APTT)、凝血酶原时间(PT)、凝血酶时间(TT);每组重复3次。②动静脉分流模型:健康广西巴马小香猪1只制作猪动静脉分流模型,分为银杏内酯B药物支架与316裸金属支架2组,采取相应干预;每组重复3个样本,电子放大镜下观察支架表面血栓形成的情况。③高负荷血栓模型:健康广西巴马小香猪2只制备高负荷血栓模型,分为银杏内酯B药物支架与316裸金属支架2组,采取相应干预;每组重复3个样本,观察2组支架形成支架内血栓的时间。 结果:①体外抗凝血试验结果显示,银杏内酯B涂层组APTT时间、TT时间较316L不锈钢组明显延长。②动静脉分流模型研究结果显示,银杏内酯B药物洗脱支架血栓重量较316L裸金属支架支架血栓重量少。③高负荷血栓模型研究结果显示,银杏内酯B药物洗脱支架平均在(140.7±12.3)s形成支架内血栓,而对应316L裸支架平均在(52.7±6.0)s形成支架内血栓,差异有统计学意义(P<0.05)。 结论:银杏内酯B药物洗脱支架具有良好的抗血栓作用,有望成为药物洗脱支架的涂层药物。
Objective: To observe the anti-thrombus function of ginkgolide B drug eluting stent in models. Methods ① Detection of anticoagulation effect in vitro:2 ml sodium citrate anticoagulation was dripped into tubes. We will take ginkgolide B and 316L bare metal stents for comparison. The APTT, PT, and TT were detected in each group for 3 times. ② Arteriovenous shunt models:a healthy pig was used to establish the model and divided into two groups:ginkgolide B stent group and 316L bare metal stent group. The thrombosis in each group was observed by electric magnifier. ③ Model of high load thrombosis:two healthy pigs were used to establish the models and divided into ginkgolide B stent group and 316L bare metal stent group, each group was treated with corresponding measures. The time of thrombosis in each group was observed. Results: ① APTT time and TT time were significantly prolonged in ginkgolide B coating group in vitro. ② Results in arteriovenous shunt thrombosis models showed that the weight of stent thrombosis was lighter in Ginkgolide B drug eluting stent compared with 316 L bare metal. ③ Results in high load thrombosis models showed that the ginkgolide B drug eluting stent in an average of (140.7±12.3) seconds stent thrombosis, and corresponding 316L stents an average of (52.7±6) seconds stent thrombosis, with a statistical difference between the two groups (P<0.05). Conclusion: Ginkgolide B drug eluting stent has good anti thrombosis effect, which is expected to be a coating drug in eluting stent selection.
