目的

观察针刺对失眠大鼠基底前脑星形胶质细胞相关标志物的影响,探索改善失眠的针刺治疗作用机制。
方法

将60只SD大鼠随机分为空白组、模型组、假针刺组、针刺组、西药组。腹腔注射对氯苯丙氨酸(PCPA)复制失眠模型,每组给予对应干预措施。采用戊巴比妥钠翻正实验检测睡眠时间和睡眠潜伏期;酶联免疫吸附分析(ELISA)检测血浆中胶质原纤维酸性蛋白(GFAP)、S100钙结合蛋白B(S100B)、脑源性神经营养因子(BDNF)、海马胶质细胞源性神经营养因子(GDNF)含量;免疫组织化学染色法检测大鼠基底前脑GFAP、S100B蛋白阳性表达。
结果

①PCPA造模后,大鼠呈现睡眠潜伏期延长,睡眠时间缩短(

0.01)的表现。针刺治疗后显著缩短了睡眠潜伏期,显著延长了睡眠时间(

0.01)。②与空白组比较,模型组与假针刺组血浆GFAP、S100B含量及基底前脑GFAP、S100B阳性表达均有上升(

针刺可改善睡眠状态,使睡眠潜伏期缩短,睡眠时间延长,可能是通过调节PCPA失眠大鼠基底前脑星形胶质细胞相关标志物蛋白表达实现的。
Objective

To observe the effect of acupuncture on astrocyte-associated markers in the basal forebrain of insomnia-model rats and explore its therapeutic mechanism in improving insomnia.
Methods

Sixty Sprague-Dawley rats were randomly divided into five experimental groups: blank control (BC), model (M), sham acupuncture (SA), acupuncture (A), and Western medication (WM). Insomnia was induced via intraperitoneal injection of p-chlorophenylalanine (PCPA). Interventions were administered accordingly. Sleep duration and latency were assessed using a pentobarbital sodium-induced righting reflex test. Plasma levels of glial fibrillary acidic protein (GFAP), S100 calcium-binding protein B (S100B), brain-derived neurotrophic factor (BDNF), and hippocampal glial cell line-derived neurotrophic factor (GDNF) were measured by enzyme-linked immunosorbent assay(ELISA). GFAP and S100B protein expression in the basal forebrain was examined using immunohistochemical analysis.

① Post-modeling, M group rats exhibited significantly prolonged sleep latency and shortened sleep duration compared with the BC group (

Conclusions

Acupuncture improves sleep by shortening latency and extending duration. This effect may be mediated through modulating the expression of astrocyte-associated markers (GFAP, S100B, BDNF, GDNF) in the basal forebrain of PCPA-induced insomnia rats.
